The aim of the present research was to develop and the evaluate microemulsion gel-based for buccal delivery of Prednisolone (PRL). Microemulsion will be provided stable system with small particle size that enhanced drug solubility and then be converted into solid nano-scale nanoparticles by using freeze drying process. The solubility of PRL in various surfactants on oleic acid oil based was evaluated. Characterizations of the prepared buccal tablets were performed carefully and observed visually. Drug delivery through buccal based microemulsion is promising areas to achieve control release with enhances bioavailability. The qualities of the microemulsion were depended on the concentration of oil, surfactants and water. Also, particle size, clarity, conductivity and pH value were also important parameters that strongly affect the structure of the microemulsion system. The qualities of microemulsion are affecting the quality of buccal too. The combination of carbopol and mannitol was a critical factor in forming desired buccal tablets through freeze drying process. Thus, the effect of carbopol and its concentration on buccal qualities, i.e. physical appearance, mucoadhesive strength, swelling and hydration, in vitro drug release and ex vivo mucoadhesive retention time were studied.
