dc.description.abstractThe aim of this study is to examine the effect of different porcine follicle size to the developmental meiotic competent after in vitro maturation (IVM). Ovaries were collected from local adult and non-pregnant slaughtered pig. Porcine oocytes were classified into three different groups, which depended on follicle diameter of 3-4 mm (Group1), 4-5mm (Group2), 5-6mm (Group3), and collected using dissection method. These groups were cultured in in vitro maturation for 40-42 hours, at 37.5oC, in 5% CO2 air incubator. After maturation process, oocytes were examined with the cumulus expansion rate before being denuded of cumulus around and fix in aceto alcohol. These specimens were kept from 7-10 days before staining in aceto-orcein 1% to obtain the proportion of matured oocytes over immature ones. With all harvested data, it is indicated that oocytes from Group3 (68.19%) and group 2 (52.12%) have a significant difference and acquire higher percentage of meiotic competent compared with Group1 (7.15%). Therefore, it is the most effective way to use oocytes from follicles of 4-6 mm in diameter, especially 5-6mm, for IVM process in experimental research and subsequent technology. Meanwhile, the oocytes from group 1 (follicle size 3-4mm) are needed to pre-incubate them in IVG for a time before introducing to IVM process.
