dc.description.abstractOsmotic stresses, especially drought, are the major stress factors that affect yield and stability of crop production. Therefore, scientists have been trying to understand and dissect the mechanisms of how plants tolerance to drought using a variety of approaches; however, success has been limited. According to previous study GmHP08 gene has displayed high level expression in the root and shoot under both normal and dehydration conditions. In this study, we amplified, cloned and sequenced GmHP08 gene from local soybean DT51 cultivar. The binary vector generated pBI121 containing GmHP08 gene construct was introduced into E.coli DH5α strain then Agrobacterium tumefaciens (A. tumefaciens) EHA101 strain by heat shock and freeze-thaw method, respectively. As result we succeeded to design recombinant vector carrying GmHP08 gene for further in planta study on mechanism of GmHP08 gene involve in drought stress. For further, this construct can be used in genetic engineering to improve the drought tolerance of soybean and other crops.
