dc.description.abstractFully grown oocytes-derived from large follicle (4-8mm in diameter) obtain full meiotic competence and reach to metaphase II (MII) after culturing in invitro maturation medium. On the other hand, the growing oocyte with small antral follicle (2-3mm in diameter) would be difficult to be mature if they are only cultured in in-vitro maturation medium. Hence, the pre in-vitro maturation system designed to support growing oocyte can develop and give a higher chance to reach metaphase II when they were cultured in in vitro maturation. In this experiment, the potential optimal pre in-vitro maturation durations were 6 hours, 8 hours and 10 hours for growing oocytes collected from small antral follicle (2-3mm in diameter). Then, the three groups were followed by in vitro maturation for 22 hours and their proportion of successful oocytes at metaphase II were examined. The result showed that the growing oocyte with 6h pre-in vitro maturation culture gave the highest percentage of oocytes at Metaphase II (72.72%) among the three groups. The morphology of the nuclear chromosome was recorded, which was almost at stringy chromatin (SC) and germinal vesicle I (GVI) stage (39.21% and 31.37%, respectively). As the result, for the small antral follicle (2-3 mm in diameter), 6 hours culture in pre in-vitro maturation medium and 22 hours in invitro maturation culture were the best period of time for them to reach metaphase II stage and extruded 1st polar body as well as achieved the highest oocyte meiotic competence in the culture condition.
